In AD5144R Datasheet, command 1(in table 14) is don't care.
And I have quetion.
If we send this command, Is ACK Hi/Low?
I think you mean command 0, no operation.
Any command received will be ACK, but in this particular command, the part is not going to do anything internally.

Thats may no sense... Could you provide some plots fo the digital transmission? Im specially interested in the I2C address frame and first frame.
I2C address is 01010000 00000001 XXXXXXXX.(AD5144A)
But this command is not for AD5144A and is for other IC in same slave address.
I worry about some malfunction.
By the way , I have AD5524 evaluation board and I tried to evaluate NOP condition.
I use ADI software in homepage.
And this software have NOP button.
Below attached file is result.
After NOP address , this IC respond ACK.
Is this Hi-Z?
To answer your original question, the ACK is Low.
I did a quick test on the device and I am attaching a Screen shot of the I2C lines for Command 0 from Table14 in datasheet of AD5144A:
Device address : 0101100 (Table 13)
Control bits : 00000110
Data bit : 00110100
(ADDR0: Vlogic; ADDR1: GND)
You can notice that at every 9th Clock is ACK is received by pulling the SDA line LOW.

If this IC didn't have ack or nack responds, I would consider whether there are some malfunction or not.
You cannot entirely base the IC is malfunctioning based on the ACK or NACK response. Please check the following item:
1. What is the logic levels in ADDR0 and ADDR1 pin? Is the address that you are sending is matching the Value in Table.13?
2. Is there any devices which has the same address as your digipot? If so you need to change the address.
3. If you are using dual-supply the logic supply should not be greater than VDD.
