I was wondering, if I have an ADC chip and I want to output to both a DAC chip and just have an I2S output to a connector, can I just tap off the same I2S line to both a DAC and a connector?

Yes, you can do this but you have to give some attention to several signal integrity issues.
1) Length of the traces
2) The stub out to a connector will cause a reflection and RFI emissions.
3) The length and type of wires used when something is connected to this connector.
4) Proper termination of the lines.
There are many ways to deal with this. You may want to consider using a buffer to drive this external line. You can get fancy and have an enable for this buffer so you can shut it off.
Just using I2S at 48kHz is not too difficult but going to higher sample rates and certainly, TDM format will make this more difficult but it certainly can be done.
