Is there a property of the Digital Isolator that makes necessary the 4/0, 3/1, 2/2 configurations instead of just rerouting my signals with a 4/0?

There are several reasons why rerouting signals is not desirable or even possible.
1) You can not reroute your signals across the isolation barrier, the channels are not dependant and share grounds and VDD on each side of the part which would circumvent the isolation.
2) Rerouting channels to the other side of a part is not possible because it violates creepage and clearnce requirements.
Optocouplers find it very hard to mix channel directions because of their manufacturing process, they would have mixed direction isolators if they could do it economically. So Optocouplers have a lot of singles and duals so the designer can flip them around one or two at a time.
