We will use the AD7195 for a single channel (AIN1 - AIN2). All other will not be used (AIN3, AIN4 and AIN COM).

Unused pins can be left floating. However, it is good practice to tie unused inputs to a known voltage. So, I suggest connecting the pins to AGND.

I want to make sure it. I'm using AD7190, when in buffered mode, the input ranges is "AGND + 250 mV and AVDD - 250 mV". So for unused inputs, I can omit this rule? Won't it affect the channel I'm using?
Apologies I don't get the question. When you enabled the buffer in configuration mode, the input ranges at selected channel should be "AGND + 250 mV and AVDD - 250 mV". So if you have unused inputs/not selected inputs you can just leave this floating.
What about tying the unused channels to ground? Will it affect the channel I used/slected?
Yes, you can connect the unused input channels to AGND.
