Are you looking to use a microphone to pick up vibrations in bone, such as through the skull bone? For this sort of application, you would want to have the microphone's sound port tightly-coupled (acoustically) to the vibrations in the bone. The microphone's sound port should probably be sealed against the skin, probably surrounded with a small gasket. The gasket will slightly increase the volume of air in front of the microphone and will probably provide a better-matched acoustic impedance at the boundary, compared to the microphone pressed against the skin with no air cavity.
