The dielectric constant does affect the speed of the signal on the line, but for typical dielectric constants, the difference will be very slight. The traces should be fabricated to be 50 ohms in Itera MT40, and the same physical lengths can be used. The TL1 and TL2 lines should be considered for their phase shift, which is proportional to their length, rather than as inductors.
