Can MAX77501 output negative voltage?

MAX77501 can't output negative voltage, because the structure is a boost converter topology. It can only do voltages above the input voltage. Its minimum output voltage is 10V.
Does MAX77501 support negative output voltage?

No. It's single-ended output based on boost DC-DC architecture. One end of the load ceramic must be connected to GND.
How can MAX77501 output random waveform?

In most cases, customer’s waveform input to MAX77501 does not start from 10V(wave trough) but from 50V(balance point). So if the input starts from 50V, will some POP noise heard or felt? Or some other issue?

If there is a sharp transition from 10V to 50V a ‘pop’ sound will occur on the piezo. This is why it is recommended that all haptic waveforms start at 10V. If the waveform has to start at 50V, it's recommended…
Is MAX77501 SPI robust enough to accpect 16MHz clock? Does it affect the sample rate of the waveform?

MAX77501 could support up to 25MHz SPI frequency, so 16MHz is ok. The waveform sample rate is independent to SPI frequency.
MAX77501 has no analog input for normal operation. Is analog input supported for testing purpose to check if 40V signal is powerful enough for their application?
