Im using petalinux with master meta-adi from 15/nov/2021, Linux version 5.10.0-xilinx-v2021.1
libiio is included in meta adi layer, but i had to add
EXTRA_OECMAKE += "-DWITH_EXAMPLES=ON"
inside "petalinux_folder/components/yocto/layers/meta-openembedded/meta-oe/recipes-support/libiio/libiio_git.bb" in order for petalinux to detect the library

The "Permission denied" error occurs when you don't have enough access rights. Running as "root" should definitely work, though.
Could you try this, as root:

echo 1 > /sys/bus/iio/devices/iio:device4/buffer/enable
echo 0 > /sys/bus/iio/devices/iio:device4/buffer/enable
cat /sys/bus/iio/devices/iio_sysfs_trigger/add_trigger
The first three commands shouldn't cause any error. The last one should return an error, but not "permission denied".
